چکیده

A low molecular weight, heat-stable factor has been purified from Euglena gracilis supernatant fraction by employing gel filtration, cation and anion exchange and paper chromatography. This endogenous compound stimulates porphobilinogenase (PBG-ase) (EC 4.3.1.8) activity, an enzyme of the porphyrin biosynthetic pathway. 1CT M folic acid and ICI" M 6-biopterin produced a significant activation, equivalent to 2—4 units of the purified factor. Elution patterns from the columns and fluorescence and UV absorption peaks suggest that this compound is a pteridine. This conclusion is further supported by the fact that both, folic acid and 6-biopterin can replace the action of the isolated factor on PBG-ase. The mechanism of stimulation is discussed.

Biosynthesis of Porphyrinogens in Etiolated Euglena gracilis Z. II. Identification of a Regulatory Pteridine

A low molecular weight compound which stimulates the synthesis of porphyrinogens from PBG was isolated and purified from etiolated Euglena gracilis Z. Absorption and fluorescence excitation and emission spectra, as well as the molecular weight, revealed its structure as an unconjugated pteridine. FTIR bands and 'H NMR signals of the purified compound were identical with those of an authentic sa...

A Comparative Study of Porphyrin Biosynthesis

LTHOUGH the primary precursors, the intermediates and most of the enzymes involved ir the biosynthetic chain of porphyrins are now known,’,2’3 knowledge of the tissue sites( s) of porphyrin formation is incomplete. Several earlier studies4’5 demonstrated that avian nucleated erythrocytes as well as rabbit bone marrow are active in this respect. Both tissues utilize either glycine and acetate ( ...

Mevalonic acid kinase in Euglena gracilis.

The isolation and partial purification of mevalonic acid kinase from Euglena gracilis is described. The product of the reaction MVA-5-P has been characterized by paper chromatography. The apparent Km values for l-mevalonic acid, ATP, and Mg(2+) are 3 x 10(-5)m, 6 x 10(-3)m, and 9 x 10(-3)m, respectively. A concentration of 1 x 10(-3)mp-hydroxymercuribenzoate completely inactivates the enzyme. A...
